Overview

Mazdutide is a weekly shot that acts on both the GLP-1 and glucagon receptors. It was approved in China in June 2025 for weight management, the first drug of its type approved anywhere. It is not approved in the United States.

How it works

Established in trials as a dual GLP-1 and glucagon receptor agonist. GLP-1 action reduces appetite and improves blood sugar; glucagon action is proposed to increase energy use and reduce liver fat. In GLORY-1 people with high liver fat saw large liver-fat reductions.

What research shows

  • GLORY-1 phase 3 RCT in Chinese adults with overweight or obesity (n=610, 48 weeks, NEJM 2025): weight change −11.0% (4 mg) and −14.0% (6 mg) vs +0.3% placebo.
  • In GLORY-1, 49.5% on 6 mg lost at least 15% of body weight vs 2.0% on placebo.
  • Participants with high baseline liver fat had liver-fat reductions of about 66–80% vs 5% on placebo (company report).

Doses are from the GLORY-1 phase 3 trial; the approved Chinese label dosing was not verified from a primary source.

Read this first: these are the amounts given to participants in published studies or written on official prescribing labels, reported for reference. They are not recommendations. Trial doses were chosen for specific patients under medical supervision, and a dose that is safe in one person can be harmful in another.

Vial math

Many research vials hold 5 or 10 mg of powder. This table shows how much ends up in each mark on a U-100 insulin syringe for common amounts of added liquid. It is arithmetic only, not a dose.

Safety

  • Most common side effects were gastrointestinal, mostly mild to moderate (GLORY-1).
  • Discontinuation due to side effects was low in GLORY-1: 1.5% (4 mg), 0.5% (6 mg), 1.0% (placebo).
  • Trial data so far are mainly in Chinese adults; long-term and cardiovascular safety are not established.

History

Licensed by Innovent Biologics from Eli Lilly for development in China. The phase 3 GLORY-1 trial was published in NEJM in June 2025. China's NMPA approved mazdutide for chronic weight management in June 2025 (announced June 27, 2025), making it the first approved dual GCG/GLP-1 agonist.

Common questions

Is mazdutide approved anywhere?

Not in the US, but China approved it in June 2025 for chronic weight management, the first drug of its type approved anywhere 2. The Chinese approval covers adults with a BMI of 28 or more, or 24 or more with a weight-related condition 2.

What does the research show?

In the GLORY-1 trial (610 Chinese adults, 48 weeks), weight fell 11.0% on 4 mg and 14.0% on 6 mg, vs a 0.3% gain on placebo 1. Nearly half (49.5%) on 6 mg lost at least 15% of body weight 1.

Does mazdutide affect liver fat?

The company reported that GLORY-1 participants with high liver fat saw reductions of about 66% to 80% vs 5% on placebo 2.

What doses were studied?

GLORY-1 tested 4 mg or 6 mg once weekly by injection for 48 weeks 1. The approved Chinese label dosing was not verified from a primary source.

What are the side effects?

Stomach side effects were most common and mostly mild to moderate 1. Few people stopped because of them: 1.5% on 4 mg, 0.5% on 6 mg and 1.0% on placebo 1.
